A New Set of Amino Acid Descriptors for the Development of Quantitative Sequence-Activity Modelings of HLA-A*0201 Restrictive CTL Epitopes

چکیده
A new set of amino acid descriptors, SZOTT (Scores of Zero dimension, One dimension, Two dimension, Three dimension) was derived from principal component analysis on a matrix of 1369 descriptors including 0D, 1D, 2D and 3D information of 20 coded amino acids. SZOTT scales were then employed to represent structures of HLA-A*0201 restrictive CTL epitopes, and further, quantitative sequence-activity modelings (QSAMs) were constructed based on partial least squares and back-propagation artificial neural networks. Satisfying results show that structural information related to biological activities of HLAA*0201 restrictive CTL epitopes can be preferably represented by SZOTT scales, which may be a useful structural representation technique for study of QSAMs of peptides.

Identification of Mycobacterium tuberculosis CTL Epitopes Restricted by HLA-A*0201 in HHD Mice
CD8+ T cells are thought to play an important role in protective immunity to tuberculosis. The major histocompatibility complex class I subtype HLA-A*0201 is one of the most prevalent class I alleles, with a frequency of over 30% in most populations. HLA-A*0201 transgenic, H-2Db/mouse beta2-microglobulin double-knockout mice (HHD) which express human HLA-A*0201 but no mouse class I, was shown t...
متن کاملMolecular Identification of Pre-Existing Immunityin Human against H9N2 Influenza Viruses Using HLA-A*0201 Binding Peptides
Background and Aims: The contribution genetic and antigenic diversity of H9N2 influenza viruses in evading from immune responses, cytotoxic T lymphocytes (CTL) epitopes in hemagglutinin (HA) protein restricted by HLA binding peptides was identified. Materials and Methods: Phylogenetic analyses were carried out for all of full length HA and deduced amino acid sequences of H9N2 viruses available ...
متن کاملEnhanced immune activity of cytotoxic T-lymphocyte epitope analogs derived from positional scanning synthetic combinatorial libraries.
The pp65(495-503) cytotoxic T-lymphocyte (CTL) epitope from cytomegalovirus (CMV) is universally recognized among CMV+ individuals who express an allele of the human leukocyte antigen A (HLA-A*0201). The relative binding affinity of the epitope to HLA-A*0201 is moderate, and its increased activity might prove beneficial in its use as a CTL epitope vaccine. A new approach to enhance the activity...
متن کاملRecognition of variant HIV-1 epitopes from diverse viral subtypes by vaccine-induced CTL.
Recognition by CD8(+) T lymphocytes (CTL) of epitopes that are derived from conserved gene products, such as Gag and Pol, is well documented and conceptually supports the development of epitope-based vaccines for use against diverse HIV-1 subtypes. However, many CTL epitopes from highly conserved regions within the HIV-1 genome are highly variable, when assessed by comparison of amino acid sequ...
متن کاملIdentification of three HLA-A*0201-restricted cytotoxic T cell epitopes in the cytomegalovirus protein pp65 that are conserved between eight strains of the virus.
The Ag specificity of the CTL response against CMV is directed almost entirely to a single CMV tegument protein, the phosphoprotein pp65. We report the identification of three peptides derived from the protein pp65 that displayed a high or intermediate binding to HLA-A*0201 molecules, which were also able to induce an in vitro CTL response in peripheral blood lymphocytes from CMV seropositive i...
